Your chance to overdose is dependant only on the drug you take, and has nothing to do with your happiness or how regularly you use it.
To add on to what Kayla said, an overdose is completely random - it's just a percent chance every time you take a drug.
I believe Ecstasy is around ~5% chance to overdose.
To add onto what Kayla and Stewie have said, every drug seems to have a different OD chance - Ecstacy seems to have a higher OD rate than Xanax, for example.
To add onto what Triplicant said, cannabis has the lowest OD chance of all, to the point where there is a merit for ODing on it with Extraordinary rarity.
In contrast, the chance of overdosing on Xanax is approximately 2%, I believe. :)
