how to increase my stats (such as manual labor, endurance intelligence and the basic stats like strength etc) and how to unlock my 2nd mission after finishing the george mission

your job does that along with allot of education (7 years with no reductions > most use merits here first ) now that you are at level 3 a company might be the another way to improve them,

To increase your stats, visit the gym and spend energy on your attack, defence, speed and dexterity. As you train more, you will begin to unlock more gyms that will give you better gains each time you train.
For job stats, you can either begin with one of the Torn starter jobs, or you can join a player-owned company. You can find player-owned companies through the Company Recruitment forum, or classified ads in the newspaper. Completing education courses (in the education tab) will also add to your job stats.
Regarding your mission question, I assume you mean that you completed a George mission and would like to begin the next one? If so, you may want to return to the Missions page and make sure you have completed it. If you're referring to the Duke missions, once those are unlocked, you will get a new one each day (up to a maximum of 3 uncompleted missions).
