Do not pay too much attention to what Duke says, just read the actual mission requirements, they are usually easy to understand.
Safe Codes are unique to your mission. Sometimes it takes a few interrogations for your target to reveal the code.
When defeating your target you get 4 options with this mission. Leave, mug, hospitalize and interrogate. Click interrogate and you have a chance that he will give you the code that you need to open Duke his safe (which Duke will send you after accepting the mission. Check your inventory). It's a 50-50 chance so he might not give code in which case you have to attack (and interrogate) the target again. If you're unlucky you need a lot of attacks to get the code. Good luck.
